EFFECT OF DIFFERENT CARBON AND NITROGEN SOURCES ON PRODUCTION OF THE SPOROTHRIX INSECTORUM ON LIQUID AEROBIC FERMENTATION

ABSTRACT

The aim of this study was to evaluated the effect of concentration of nitrogen and carbon for production of Sporothrix insectorum on liquid aerobic fermentation with alternative media. Casamino acid and dextrose were used to evaluate for concentrations of nitrogen and carbon, using different combinations: 60, 40, 20 and 10 g/L water. Evaluations were made of biomass (g), blastospores and hyphae on day 2, 4, 6 and 8. After the determination of better concentrations of nitrogen and carbon for the production of S. insectorum, evaluated alternative media with same concentrations the protein and carbohydrate: beer yeast, powder milk, soybean protein, bean, soybean milk and yeast extract + sucrose, with 60 g nitrogen/L and 20 g carbon/L. S. insectorum needs 6% of nitrogen for better development. The bean (100 g/L), yeast extract (10 g/L) + sucrose (20 g/ L), casamino acid mediums (60 g/L) + Dextrose (20 g/L), Powder Milk (200 g/L) and Beer Yeast (164 g/L) produced more than 7 x 109 blastospores and propagates/mL and a good biomass in 8 days. The beer yeast (164 g/L) is the best for production of S. insectorum because its cost is R$ 1,35/L.
