STUDY OF REPLICATION CYCLE OF INFECTIOUS BURSAL DISEASE VIRUS IN RK-13 CELL LINE

ABSTRACT

The aim of the present work was to study the replication cycle of infectious bursal disease virus (IBDV) in RK-13 cell line through presenting of cytopathic effect, viral titration and detection of viral RNA by polyacrylamide gel electrophoresis (PAGE). One field sample (BAB sample) and the vaccine strain GBV were used. The detection of extra-cellular viruses occurred before that of intra-cellular viruses, at 6 and 10 hours post-inoculation (pi) respectively. The extra-cellular viral titers were higher than intra-cellular viral titers for both samples. The cytopathic effect began at 10 hours pi for the BAB sample and 24 hours pi for the GBV sample together with the detection of viral RNA by PAGE. Comparing with literature data the replication cycle in RK-13 cell line was slower than in primary chicken embryo fibroblast cultures but equal to or faster than that in other cell lines.
