Different concentrations of the ms medium, nitrogen and sucrose on micropropagation of chrysanthemum

The effect of MS medium, nitrogen and sucrose war investigated on "in vitro" propagation of shoot nodal explants of chrysanthemum (Dendranthema grandiflora Tzvelev.) cultivars Orange Reagen. MS medium was tested using the following concentrations: 0, 25, 50, 75, 100, 125 and 150% in combination with sucrose doses of: 0, 0.75, 1.5, 3.0, 6.0 and 12.0%. Nitrogen was tested using the concentrations of 0, 25, 50, 75, 100, 125 and 150% in combination with the sucrose doses used as above. Best shoot development was observed when nodal segments were cultivated on medium containing concentrations between 50 and 150% of MS in combination with 6% sucrose. No satisfactory shoot length was observed when 50% MS was used. Roots were not observed at concentrations above 100% of MS and nitrogen. In order to obtain successful "in vitro" propagation of chrysanthemum, our results suggest the use of 75% MS, with 75% of nitrogen and 6% of sucrose.
